چکیده
Materials & methods After isolation, the CD34+ CB stem cells were cultured using a two-step protocol. First, they were expanded using a combination of (growth) factors (Flt3L, SCF, IL-3 and IL-6). Next, the cells were differentiated towards DCs for one week using medium containing Flt3L, SCF, GM-CSF, IL-4 and human serum followed by a CYTOMIX (IL-1b, IL-6, TNF-a and PGE2)-induced maturation for the last 24 hours. Finally, the CB-DC culture was electroporated with WT1-mRNA and their phenotype (cell surface markers) and function (migration and antigen presentation) were assessed.

Comparison of Functional Competence of Umbilical Cord and Adult Peripheral Blood Dendritic Cells in Allogenic Mixed Leukocyte Reaction
Background: Dendritic cells (DCs) are the most potent stimulators of primary T cell responses and play a key role in immune reactions after stem cell transplantation. Very little is known about the cord blood (CB) dendritic cells and their potential involvement in the low incidence and lower severity of acute graft-versus-host disease after CB transplantation. Objectives: The aim of this stud...
متن کاملImmediate Exposure to TNF-α Activates Dendritic Cells Derived from Non-Purified Cord Blood Mononuclear Cells
Background: Tumor necrosis factor alpha (TNF-α) is a primary mediator of immune regulation and might be required in the early stages of DC development from CD34+ cells. However, details of optimal timing of exposure to TNF-α in DC development process in monocytes or non-purified hematopoitic cells are still lacking and clear benefits of this approach to the development of DCs remain to be valid...
متن کاملOnm-19: The Role of Cord Blood Preservationin Cell Therapy
s:3604:"During pregnancy, the placenta delivers "cord blood" to the baby through the umbilical cord serving as a lifeline of nourishment from the mother to baby. At birth, "cord blood" remains in the umbilical cord and placenta and until recently, had typically been discarded. The tragedy of this practice is that "cord blood" contains very special cells called "stem cells". Recent advances in m...
متن کاملCD26+ Cord Blood Mononuclear Cells Significantly Produce B, T, and NK Cells
Background: Umbilical cord blood (UCB) is an alternative source of hematopoietic stem cell transplantation (HSCT), used in Leukemia treatment. CD26+ cells, a fraction of CD34 positive cells, are a major population of UCB cells which negatively regulate the in vivo homing and engraftment of HSCs. CD26 is highly expressed in various cells such as HSCs, immune cells, fibroblasts, and epithelial ce...
متن کاملEngineered dendritic cells from cord blood and adult blood accelerate effector T cell immune reconstitution against HCMV
Human cytomegalovirus (HCMV) harmfully impacts survival after peripheral blood hematopoietic stem cell transplantation (PB-HSCT). Delayed immune reconstitution after cord blood (CB)-HSCT leads to even higher HCMV-related morbidity and mortality. Towards a feasible dendritic cell therapy to accelerate de novo immunity against HCMV, we validated a tricistronic integrase-defective lentiviral vecto...
